Nike commissioned 45:33 for its Nike+ Original Run series, and it was first released through the Nike Music Store on iTunes on 17 October 2006. Murphy recorded it between July and September 2006 at DFA Studios. The claim that it was refined over treadmill runs was, Murphy later said, entirely a lie, and he does not jog. What he wanted, he said, was a long-form record like Manuel Gottsching's E2-E4, and the artwork echoes that record. The title is not the running time: it refers to the two common vinyl speeds, 45 and 33 RPM.

Parts of it were later used on Sound of Silver, most clearly Someone Great, which appears on 45:33 as an instrumental section later numbered as track 3. DFA reissued it on CD and vinyl on 12 November 2007 with three bonus tracks, Freak Out/Starry Eyes, Murphy and Eric Broucek's dub of North American Scum and Hippie Priest Bum-out. Those three also came out together on the Confuse the Marketplace EP on 11 December 2007. Metacritic scored it 86. Gottsching himself was critical, calling it a megamix of his work.

Pitchfork called it an original workout mix pieced together like a DJ mix. At the final concerts at Terminal 5 and Madison Square Garden the band played it in full in the second set, with Sound of Silver standing in for Part 3 and Part 3 held back for Someone Great. The digital release of the farewell show titles its parts 45:33 Intro, You Can't Hide / Shame On You, Out in Space and Ships Talking.
