The band's first single, released on 11 August 2003 from Youth and Young Manhood, reached number 23 on the UK Singles Chart. The same recording is on the Holy Roller Novocaine EP and on the album. The title comes from a line in Thin Lizzy's version of the Irish folk song Whiskey in the Jar, where Molly's chambers is archaic slang for a brothel. Nathan Followill said it was one of four songs the band had ready after a month in a basement. It appeared in a Volkswagen Jetta commercial, and with California Waiting and Holy Roller Novocaine in the Farrelly brothers film Stuck on You.

An alternate take called Molly's Hangover is slower until near the end and has a different style of backing vocals and guitar solos. A 10-inch vinyl edition, limited to 6,000 copies, carried a live Holy Roller Novocaine recorded at the Birmingham Academy on 27 June 2003.
